Antisense oligonucleotide (ASO) is a single-stranded DNA or hybrid oligonucleotide, usually 15-25 bases long, which can hybridize with the target RNA sequence by its base sequence arrangement to mediate the cleavage of RNA by RNase H, thereby inhibiting the function of non-coding RNA (such as miRNA, siRNA, piRNA, snoRNA, snRNA, exRNA, scaRNA and lncRNA) or blocking the protein translation of mRNA. In order to improve nuclease resistance, the introduction of phosphorothioate (Phosphorothioate, PS) bonds in the backbone sequence can be used to improve nuclease resistance, thereby enhancing intracellular stability. It has the ability to interfere with both cytoplasmic and nuclear RNA, and can be directly used in in vivo experiments.
