In living organisms, gene silencing caused by siRNA, miRNA, and ASO-mediated RNA interference (RNAi) is an important form of gene expression regulation that specifically initiates target mRNA degradation or protein translation expression inhibition, thereby reducing gene expression and function.

Since RNAi targets are upstream mRNA, lncRNA, circRNA, miRNA and other RNA molecules, and there is a diversity of upstream and downstream gene action mechanisms, RNAi has the advantage of being able to achieve many small molecules and monoclonal antibodies that cannot become drugs, such as targets with complex or unclear protein structures, targets with shear mutations, lncRNA, circRNA, miRNA-regulated targets, etc. Currently, hundreds of RNAi drugs have entered clinical studies worldwide, and several siRNAs have been approved, which will lead the wave of new drug development in the next 10 years.
