Uracil N-Glycosylase (UNG)
UNG is often used to eliminate residual contamination in PCR amplification. When PCR reaction solution is prepared, dUTP and dNTP are mixed in a certain ratio (or dUTP is used instead of dTTP) so that both amplification products contain deoxyuracil. The residual contamination of previous PCR products can be eliminated by adding 0.2-1 U UNG to the PCR mix and adding a 2-10 min 37°C holding step before performing PCR amplification, and UNG can be inactivated during the pre-denaturing heating at 95°C for 2 min in the PCR cycle.
