The assay is based on the TaqMan principle and relies on the 5’→3’ exonuclease activity of Taq polymerase as well as the dual labeling of the probes with fluorescents and quenchers. During PCR the Taq polymerase cleaves and removes annealed probes, releasing the previously quenched fluorescent signal.
The target (Rat) sequence is a mitochondrial multi-copy gene. The assay is composed of two principal steps: extraction of total DNA from meat (or food) samples, and the BioZues® HALAL Real-time PCR Kit (Rat), a qualitative duplex real-time PCR test for the detection of Rat-specific genes and exogenous internal positive standards (IC) using specific primers and probes labeled with fluorescent dyes. The target sequences and IC are detected through the FAM and HEX (VIC) channel, respectively.
